Why do people age?
Starting from birth and throughout the whole life human cells constantly accumulate such structures as old defective mitochondria, peroxysomes, lysosomes and “broken defective proteins”, epigenetic mutations, free radicals triggering the processes of apoptosis (programmed cell death) and premature aging, which are commonly called cellular garbage (CG) lipofucsin.
Lipofuscin pellets are wastes from the body, its life — defective proteins, disposed and dead cells that are unable to re-engage key metabolic reactions. CG is one of the types of damage (breakdowns) at the cellular level, which prevents the normal functioning and harmonious interaction of human cells in metabolism and is the main cause of aging and numerous diseases (including cardiovascular and oncological diseases, diabetes, neurodegenerative changes and diseases of the musculoskeletal system, lungs and etc.)
Non-aging is moral and good. There is a very simple ethical logic behind this statement — if life and active longevity are the highest values and reducing the burden of suffering from disease is another value, then prolonging life by compensating age-dependent processes and diseases, improving the quality of life is as high a priority as caring for children and the elderly. Ultimately, the goal of longevity supporters is to improve quality of life and prolong life!
Lipofuscin is an autofluorescent pigment (from yellow to brown in colour) of glycolyprotein nature contained in all organs and tissues; it consists of oxidized and transversely stitched proteins and lipids, which makes it difficult to degrade and excrete it from cells. It contains phospholipids, cholesterol, triglycerides, fatty acid oxidation products and various enzymes known as amino acids. Lipids make 20-50%, proteins make 30-60% and 9-20% are iron-containing residues of black colour, which are not hydrolysed. The percentage of lipofuscin depends on the organ in which it is formed. The accumulation of lipofuscin has two stages — the so-called early pigmentation and late pigmentation. Lipofuscin in early stages is pale yellow in colour and lipofuscin in late stages already has pellets and its colour turns brown. The pigment is accumulated in lysosomes, peroxisomes and/or inside mitochondria. Physiological and pathological processes in the body influence the amount of lipofuscin.
The presence of lysosome enzymes (alkaline phosphate) in lipofuscin suggests that oxidative stress may cause the appearance of lipofuscin. The nature of the accumulation of lipofuscin in cells is associated with the destruction of cellular organelles, which have not been disposed by lysosomes.
Lipofuscin takes part in programmed cell death. In the early years of life, the impact of cellular debris is compensated by the coordinated work of free-radical oxidation enzymes, activation of the apoptosis system and hormonal regulation by the type of feedback, so, normally it almost does not appear. However, over the years, aggressive metabolic products gain the critical mass as the result of metabolism, consumption of «unhealthy» food and decreasing of activity. Therefore, a breakdown occurs and the process of accelerated aging is started. As a result of decompensating of the harmony processes of aging and anti-aging, age-dependent diseases start to develop.
The main sources of cellular garbage (CG) are the following:
- the metabolism, which even under favourable conditions “fails” from a young age, deforming and disabling the cells of the human body
- certain types of ingredients in modern food products that enter the body during the digestion processes are converted into harmful substances that destroy cells inside and out
- when cellular fragmented structures are not excreted from the body, they accumulate, lead to various dysfunctions of organs, system and diseases
Results of degenerative processes of premature aging are as follows:
Dystrophic formations that envelop the cell from the outside. They are formed from “bad” cholesterol. Its main source are the degenerative processes of protein, lipid and carbohydrate metabolism although the basic nutrients come from everyday food.

To destroy this ballast in the cells, special incinerators work, they are called lysosomes. Lysosomes act as cell cleaners and take part in the recycling of organic cell material and intracellular «digestion» of macromolecules. They are necessary for the degradation of intracellular components (organelles).
The degradation of the cellular components and macromolecules for disposal occurs in the lysosomom under the influence of special fissile enzymes, one of which is alkaline phosphatasis. Cleaning of cell fragments and damaged cells is carried out with the help of an army of macrophages, which are the immune guardians of the body. Macrophagues cope with their task well, absorbing and neutralizing dangerous substances in a healthy young body. With age or due to serious diseases, people have age-related and pathological accumulations of intracellular waste.
Lipofuscin levels as a biomarker of aging.
Lipofuscin doesn’t cause a disease. Its presence only indicates a change in the functioning of cells, it is an indicator of lysosomes and mitochondria disruption, which leads to the deterioration of cell metabolism, which in turn indicates the approaching of apoptosis and is a symptom of pathology of cells, tissues and organs in the human body.
The mitochondrial—lysosomal axis theory of aging most clearly defines the connection of lipofuscin with aging, according to which the age related accumulation of lipofuscin cluttering lysosomes is closely associated with the progressive violation of autophagy, oxidative stress and dysfunction of mitochondria.
Lipofuscin is an important sign of aging in the whole. It has been established that life expectancy decreases with the increase in accumulation of lipofuscin. Lipofuscin causes a gradual age-related accumulation of damaged mitochondria, other types of organelles and, as a result, cell dysfunction of post-mytotic cells, organ dysfunction and their aging. Lipofuscin is accumulated in the cells of the brain, heart, retina, skeletal muscles, skin and other organs. Therefore, lipofuscin is used as a biomarker of aging.
What influences accelerated accumulation of lipofuscin?
- Regular over-consumption of iron-containing foods and saturated fatty acids
- Chronic alcohol consumption
- Overactive exercise also often stimulates accelerated accumulation of lipofuscin in the liver, kidneys and skin
